Does the white have to be exactly white?

For an Amazon main image, yes — the rule is RGB 255, 255, 255, and a photograph of a white wall is never that. Padding a cutout with #FFFFFF is, because the colour is set rather than photographed.

Can I keep the transparent version too?

Download it before the resize step. A transparent PNG is what you want for your own storefront, banners and email, where the background is whatever the layout is.
